You haven't given us anything to work with other than it's a .50 caliber barrel and you want to make a rifle.
What kind of .50 cal. barrel is it? Machine gun? Blackpowder?
What type of rifle do you want to make?

Sure it can.

Depends on how you look at your needs, abilities and wallet.
There isn't an action made that you can just screw that barrel into unless it's one of the originals... You can buy an action suited to the .50 BMG cartridge and then modify the barrel to fit that action. Then you can call McMillan and order a stock big enough to handle the barreled action or design and build one yourself.
There are also instruction books written that have fairly detailed instructions with illustrations that can guide you through a build of your own. The drawback here is that you need access to the machinery and the ability/talent to make the parts. The additional materials required for the build have escalated incredibly in price so you need to have some deeper pockets than before.

In my opinion, you'll be better off selling the barrel and spending the money for something useful.
